Csm6 Type III CRISPR-Associated Proteins:

Cyclic Oligoadenylate stimulated RNAse Activity useful for Tandem RNA Detection Assays

Csm6 family proteins consist of a CARF (CRISPR associated Rossman fold) domain which binds cyclic oligoadenylate (cOA) and a HEPN (Higher
Eukaryotes and Prokaryotes Nucleotide binding) domain which possesses ribonuclease activity. The Csm6 RNA degradation activity is stimulated by cOA and functions to protect the bacterial cell from invasive genetic elements.

Combining RNA-guided Cas13 (e.g. LwCas13a or LbuCas13a) and Csm6 nucleases (Fast Integrated Nuclease Detection In Tandem: FIND-IT) enables sensitive, direct RNA detection in a one-step assay format that is amenable to point-of-care infection diagnosis as well as to a wide range of other diagnostic, therapeutic, or research applications.
