Encodes the adenine base editor ABE8e, an engineered CRISPR-Cas9 variant consisting of nSpCas9 (D10A) Nickase from Streptococcus pyogenes fused to the TadA-8e adenine deaminase, that catalyzes A•T to G•C conversions with enhanced efficiency and precision for therapeutic genome editing. This synthetic mRNA enables transient, dose-controlled delivery of ABE8e for research in:
Correcting pathogenic SNVs (e.g., Tyr402Cys in C1-INH for hereditary angioedema), developing ex vivo cell therapies (e.g., base-edited CAR-T cells), and studying epigenetic modulation via targeted adenine methylation
