    TriAltus Bio´s highly purified recombinant Lachnospiraceae bacterium Cas12a (LbCas12a, LbCpf1) protein (>90% purity) with 2 C-terminal nuclear localization signals (NLS) is used for CRISPR-based high fidelity gene editing technologies.  The enzyme was expressed in E. coli and purified by TriaAltus Bio´s proprietary CL7 Tag/CLiM technology and is delivered tag-free.

    LbCas12a protein combines with guide RNA (gRNA) to form a stable ribonucleoprotein (RNP) complex that enables the inactive nuclease to be guided to the specific double stranded DNA (dsDNA) site. Once guided to the specific cleavage site, LbCas12a becomes activated to specifically and precisely cleave the dsDNA and to indiscriminately trans-cleave single stranded DNA (ssDNA) nearby.

    TriAltus’ highly pure, tag-free LbCas12a is ideal for genome editing, CRISPR diagnostics, and multiplexed gene targeting. This recombinant enzyme, from Lachnospiraceae bacterium, is expressed in E. coli and includes two C-terminal nuclear localization signals (NLS). LbCas12a is a Class 2, Type V-A CRISPR nuclease that cuts DNA at thymine-rich PAM sites (5’-TTTV), creating staggered 5′ overhangs for precise gene insertion. It also processes its own guide RNAs, making it well-suited for editing multiple genes at once. With lower off-target activity than Cas9 and strong trans-cleavage activity, LbCas12a supports both high-fidelity genome engineering and sensitive CRISPR-based detection.

    LbCas12a shows a very good activity also at lower temperatures, and has been widely used for plant genome engineering projects.
