    Description

    Recombinant Cas9 Nickase Protein for CRISPR Knock-In Homology Directed Repair, and Prime Editing: SpCas9(H40A)

    SignalChem’s highly purified recombinant Streptococcus pyogenes Cas9(H840A) protein (>70% purity) with N-terminal nuclear localization signal (NLS) is used for CRISPR Knock-In applications employing homology directed repair by paired nicking along with target strand nickase, as well as for prime editing, and for CRISPR in vitro applications.  The enzyme was expressed in E. coli with His tag, which was used during the purification process. SpCas9(H840A) protein combines with guide RNA (gRNA) to form a stable ribonucleoprotein (RNP) complex that enables the inactive nickase to be guided to the specific target site. Once guided to the specific cleavage site, SpCas9(840A) becomes activated to specifically and precisely cleave the non-target strand.
