SignalChem’s highly purified recombinant Enterococcus italicus Csm6 (EiCsm6) protein (>95% purity) without nuclear localization signal (NLS) is used for CRISPR-based Tandem RNA detection technologies and for CRISPR-mediated in vitro applications. The enzyme was expressed in E. coli and purified using a his tag.
EiCsm6 protein consists of a CARF (CRISPR associated Rossman fold) domain which binds cyclic oligoadenylate (cOA) and a HEPN (Higher Eukaryotes and Prokaryotes Nucleotide binding) domain which possesses ribonuclease activity, stimulated by the cOA binding.
Combining RNA-guided Cas13 (e.g. LwCas13a or LbuCas13a) and EiCsm6 nuclease (Fast Integrated Nuclease Detection In Tandem: FIND-IT) enables sensitive, direct RNA detection in a one-step assay format that is amenable to point-of-care infection diagnosis as well as to a wide range of other diagnostic, therapeutic, or research applications.
