Easy-to-run, single-day scAIRR-seq assay that uses multiplex RT-PCR and NGS technology to profile paired BCR sequences at single-cell resolution
The DriverMap™ Single-cell Adaptive Immune Receptor (scAIR) Sequencing Assay enables identification of BCR paired full-length (V(D)J) immune receptor sequences of single B cells sorted in a 96-well plate. The paired full-length receptor sequence information is necessary to clone and express functional BCRs for follow-up antigen discovery studies and the development of antibody-based therapies.
• The assay works with FACS-sorted cells in a 96-well plate without needing special microfluidic equipment or complex split-pool barcoding workflows
• Simultaneously amplify—in a single multiplex RT-PCR reaction—full-length (CDR1-CDR2-CDR3) regions for all 3 BCR chains (IGH, IGL, IGK) of single B cells sorted in 96-well plate(s)
• The use of well/cell-specific barcodes and of primers which are highly specific to B-Cell Receptors and incoporate Validation Barcodes (VBC) enables a highly efficient protocol for pooled Multiplex RT-PCR based NGS library preparation
• NGS data can be analyzed with MiXCR, Platforma, and RSEM software packages available online
