Pooled Lentiviral CRISPRa sgRNA Libraries for Genome Wide CRISPR Activation Screenings
The CRISPRa platform provides an alternative genetic screening approach enabling the identification of genes whose activation initiates a response. In contrast to CRISPR Knock Out the effect is based on modulating gene activity, does not alter the underlying DNA structure, and is reversible.
Cellecta´s CRISPRa libraries make use of the engineered version of nuclease-deactivated Cas9 (dCas9) complexed with the transcriptional activator VPH. The libraries´ sgRNAs designed for CRISPRa are targeting the upstream promoter regions of 19,000 protein coding human or mouse genes and are guiding the dCas9-VPH complex to these regions for activating genes.
Pooled CRISPRa libraries enable broad based gene screens that assess the effects of repression on thousands of different genes on cell growth or other screenable phenotypes. CRISPRa pooled libraries screens can be used to identify potential therapeutic targets responsible for controlling growth and differentiation, regulating disease development, or other biological responses. The CRISPRa system is the only CRISPR-based screening approach that enables genome-wide gain-of-function screens to identify endogenous genes that change phenotypes when activated, rather than disrupted.

Cellecta Genome Wide CRISPRa Lentiviral sgRNA Libraries
- Off-the-shelf, genome-wide, lentiviral CRISPRa libraries for human and mouse
- Libraries which comprise two sgRNAs targeting each gene for stronger activation (dual-sgRNA constructs libraries) are only commercially available from Cellecta
- Both plasmid (200 ug) and pre-packaged lentiviral particle (2x 10^8 TU & 1x 10^9 TU) formats are available
Cellecta’s CRISPRa human and mouse genome-wide sgRNA libraries have been designed to target ~ 19,000 protein-coding genes using 5 sgRNAs designed against the promoter region based on Horlbeck, et al. Cellecta´s dual-guide CRISPRa and CRISPRi libraries use the same 5 effective sgRNA in dual combination on each of 5 constructs (sgRNA 1 & 2, sgRNA 1 & 3, sgRNA 2 & 3, etc.).
