Pooled CRISPRa Libraries

CRISPRa targeted gene activation provides alternative genetic screening approaches to identify genes whose activation initiate a response, a so called gain of-function screen. CRISPRa libraries that activate endogenous gene expression, make use of engineered versions of deactivated Cas9 (dCas9) which have proven to be effective modulators of gene expression when complexed with activator proteins, e.g. dCas9-VPH.
A special sgRNA design, based on Horlbeck, et al. (eLife 2016;5:e19760 DOI: 10.7554) increases dCas9-VPH activation.
Unique are Cellecta´s Dual sgRNA CRISPRa libraries which uses the same 5 effective sgRNAs in dual combination on each of 5 constructs (sgRNA 1 & 2, sgRNA 1 & 3, sgRNA 2 & 3, etc.).
Nucleus Biotech provides Cellecta´s pre-made, pooled lentiviral libraries targeting the promoter regions of all human or mouse protein-coding genes.The libraries are available as 200 ug plasmid and pre-packaged 2 x 10^8 TU and 1 x 10^9 TU lentiviral particles.
