Ribosome Profiling
Quantitative transcriptome wide measurement of ribosome occupancy by Ribo-seq
eRibo Pro: Eclipsebio’s version of ribosome profiling
eRibo Pro Ribosome profiling is an advanced service that combines RNA sequencing (RNA-Seq) with ribosome-protected fragment sequencing (RPF-Seq), also known as ribosome profiling or Ribo-Seq. This integrated approach provides a complete picture of gene expression from mRNA transcription to protein translation.
Send us your cell samples and our partner Eclipse Bio will perform a comprehensive ribosomal profiling service from sample processing to next-generation sequencing and full data analysis.

The service includes the preparation of two NGS libraries for each sample:
• An RPF library containing all mRNA regions that were actively translated at the time of sampling. After locking the ribosomal-mRNA complexes in place and digesting the free mRNA fragments, all gene sequences of the active translatome are obtained.
• An RNA-Seq library representing all mRNAs in your sample, providing information on the expression of each gene in the transcriptome
Comparative NGS and data analysis will result in a comprehensive report demonstrating changes in translation in response to cell treatments in a quantitative manner.
What you can address with ribosome profiling:
What sequences are being translated?
- You want to know which mRNAs are being translated in a cell at a specific time and which open reading frames are active?
- You would like to analyse overall translation efficiency and compare translation rates under different conditions or in different cell type?
Is a treatment affecting translation ?
- You want to study translation regulation by various factors, such as chemical substances, microRNAs, and RNA-binding proteins?
- You are interested in post-transcriptional regulation mechanisms?
Where are Ribosomes pausing?
- You want to understand the mechanism of ribosome stalling and gain insight in its role into both normal cellular processes and disease states?
- You like to study if there are differential ribosome stall sites?
What you get with Eclipsebio’s eRibo Pro ribosome profiling service:
Two NGS data sets for each provided cell sample:
- From an RPF library containing all mRNA regions that were actively translated at the time of sampling.
- From an RNA-seq library representing all mRNAs in your sample.
Ribosome occupancy analysis:
- Comparison of ribosome occupancy (RO) between two sample groups, where RO is defined as the RPF counts over the RNA-Seq library counts for each gene.
Trancriptome analysis:
- Determination of codon usage
- Detection of open reading frames (ORFs)
- Identification of stalls
Ribo Pro™ is an exclusively licensed method for profiling ribosome-associated RNAs and the transcriptome. Our service provider Eclipsebio is sole licensee of the Ribo-seq method covered by U.S. Patent Numbers 8,486,865, 9,284,547 and 9,677,068.
